Human T-Lymphoid Progenitors Generated in a Feeder-Cell-Free DL-4 Culture System Promote T-Cell Reconstitution in NOD/SCID/γc−/− Mice
Researchers described the development of a new culture system based on the immobilized Notch ligand Delta-like-4 (DL-4). Culture of human CD34+ cord blood cells in this new DL-4 system enabled the in vitro generation of large amounts of T-cell progenitor cells that displayed the phenotypic and molecular signatures of early thymic progenitors and had high T lymphopoietic potential. The Autoimmunity Associated BLK Haplotype Exhibits Cis-Regulatory Effects on mRNA and Protein Expression that Are Prominently Observed in B Cells Early in Development
The authors showed that in primary human B lymphocytes, cis-regulatory effects of disease associated SNPs in the gene B lymphocyte kinase (BLK) are restricted to naïve and transitional B cells. Cis-regulatory effects are not observed in adult B cells in later stages of differentiation. Allelic expression bias was also identified in primary human T cells from adult peripheral and umbilical cord blood, thymus and tonsil, although mRNA levels were reduced compared with B cells. Human Thymic MR1-Restricted MAIT Cells Are Innate Pathogen-Reactive Effectors that Adapt following Thymic Egress
Human mucosal-associated invariant T (MAIT) cells express the semi-invariant T-cell receptor Vα7.2 and are restricted by the major histocompatibility complex-Ib molecule MR1. The Role of WDR5 in Silencing Human Fetal Globin Gene Expression
In human bone marrow erythroid progenitor cells, levels of WDR5 were highly enriched on the γ-promoter relative to levels on other globin promoters and compared to the γ-promoter in cord blood erythroid progenitors, suggesting that WDR5 is important in the developmental globin gene expression program. The Homing of Human Cord Blood Stem Cells to Sites of Inflammation: Unfolding Mysteries of a Novel Therapeutic Paradigm for Glioblastoma Multiforme
Results indicated that multiple cytokines are involved in recruitment of human umbilical cord blood mesenchymal stem cells (hUCBSC) toward glioma stem cells, and that hUCBSC are a potential candidate for glioma therapy. [Cell Cycle] Abstract

Effect of AGM and Fetal Liver-Derived Stromal Cell Lines on Globin Expression in Adult Baboon (P. anubis) Bone Marrow-Derived Erythroid Progenitors
Adult baboon bone marrow and human cord blood CD34+ progenitors were grown in methylcellulose, liquid media, and in co-culture with stromal cell lines derived from different developmental stages in identical media supporting erythroid differentiation to examine the effect of the micro-environment on globin gene expression. Immune Recovery in Adult Patients following Myeloablative Dual Umbilical Cord Blood, Matched Sibling, and Matched Unrelated Donor Hematopoietic Cell Transplantation
Compared to HLA-identical matched sibling and unrelated donor adult hematopoietic cell transplantation recipients, quantitative lymphoid recovery in umbilical cord blood transplant recipients is slower in the first three months, but these differences disappeared by 6-12 months following transplantation.
