Study Uses Gene Editing to Take Brakes Off Lab-Based Red Blood Cell Production
Adult and cord blood stem cells treated with RNA interference produced three-to-five and five-to-seven times more red blood cells than controls, respectively. Reprogramming Human B-Cells into Induced Pluripotent Stem Cells and Its Enhancement by C/EBPα
Researchers showed for the first time that freshly isolated non-cultured human cord blood- and peripheral blood-derived CD19+CD20+ B-cells can be reprogrammed to induced pluripotent stem cells carrying complete VDJH immunoglobulin gene monoclonal rearrangements using non-integrative tetracistronic, but not monocistronic, Oct4, Sox2, Klf4 and Myc-expressing Sendai Virus. [Leukemia] Abstract

Placenta-Derived Mesenchymal Stem Cells Possess Better Immunoregulatory Properties Compared to Their Cord-Derived Counterparts–A Paired Sample Study
Investigators compared mesenchymal stem cells (MSCs) obtained from placenta (P-MSCs) and umbilical cord (C-MSCs) from the same donor, for their immunomodulatory efficacy. P-MSCs and C-MSCs showed similar morphology and phenotypic profile, but different clonogenic ability. [Sci Rep] Full Article

Different Motile Behaviors of Human Hematopoietic Stem versus Progenitor Cells at the Osteoblastic Niche
Using a xenotransplantation model and intravital imaging, scientists demonstrated that human hematopoietic stem cells (HSCs) display distinct motile behaviors to their hematopoietic progenitor cell (HPC) counterparts, and the same pattern can be found between mouse HSCs and HPCs. [Stem Cell Rep] Full Article

Increased Proportion of Hematopoietic Stem and Progenitor Cell Population in Cord Blood of Neonates Born to Mothers with Gestational Diabetes Mellitus
Scientists assessed the hematopoietic stem and progenitor cell population in the cord blood of neonates born to mothers with gestational diabetes mellitus (GDM) in a hypothesis generating pilot study, due to that neonatal polycythemia may be the consequence of GDM pregnancy. [Stem Cells Dev] Abstract

Differential Ability of MSCs Isolated from Placenta and Cord as Feeders for Supporting Ex Vivo Expansion of Umbilical Cord Blood Derived CD34+ Cells
The authors performed a systematic comparison of hematopoietic supportive capability of umbilical cord tissue- and placenta-mesenchymal stem cells (MSCs) using paired samples. [Stem Cell Res Ther] Full Article

Tumorigenic Hybrids between Mesenchymal Stem Cells and Gastric Cancer Cells Enhanced Cancer Proliferation, Migration and Stemness
Scientists investigated the role of mesenchymal stem cells (MSCs) participated cell fusion in the tumorigenesis of gastric cancer. They fused human umbilical cord MSCs with gastric cancer cells in vitro by polyethylene glycol and the hybrid cells were sorted by flow cytometer. [BMC Cancer] Full Article

Human Umbilical Cord Derived Mesenchymal Stem Cells Promote IL-17 Production from hPBMCs of Healthy Donors and Systemic Lupus Erythematosus Patients
Human umbilical cord derived mesenchymal stem cells (hUC-MSCs) were examined for their effects on lymphocytes from healthy donors and from patients with systemic lupus erythematosus. Significantly higher levels of IL-17 were produced when CD4+ T cells from healthy donors were cocultured with hUC-MSCs than that were cultured alone. [Clin Exp Immunol] Abstract

Therapeutic Effects of Umbilical Cord Blood Derived Mesenchymal Stem Cell-Conditioned Medium on Pulmonary Arterial Hypertension in Rats
As low survival rates and potential tumorigenicity of implanted cells could undermine the mesenchymal stem cell (MSC)-based therapy, researchers chose to investigate the use of conditioned medium (CM) from a culture of MSCs as a feasible alternative. CM was prepared by culturing human umbilical cord blood-derived MSCs in three-dimensional spheroids. [J Pathol Transl Med] Abstract | Full Article

Adhesion Receptors Involved in HSC and Early-B Cell Interactions with Bone Marrow Microenvironment
Investigators review recent data on adhesion mechanisms involved in hematopoietic stem cells (HSCs) and B cell progenitors interactions with surrounding bone marrow stromal cells. [Cell Mol Life Sci] Abstract

Hypoxia Regulates the Hematopoietic Stem Cell Niche
The authors focus on hypoxia-inducible factor-mediated oxygen-sensing mechanisms of adult hematopoietic stem cells and leukemic stem cells and their niche cells in the hypoxic bone marrow. [Pflugers Arch] Abstract

Spain Starts Ambitious Trial to Cure HIV Using Umbilical Cord Stem Cells
Doctors in Spain have announced the world’s first clinical trial to cure five HIV patients within three years using transplants of blood from umbilical cords. [The Local es] Press Release
